چکیده

Genertation of hydrogen by purple, non-sulfur bacteria of Rhodobacter sphaeroides O.U.001 was tested in photobioreactors made of different materials. Reactors made of ordinary sodium-type glass, borosilicate glass (Pyrex) or polycarbonate were applied in the present study. The applied medium containing malic acid and sodium glutamate inoculated with 0.11 or 0.31 g dry wt. of Rhodobacater sphaeroides was illuminated with 5 or 17 klx. Simultanous measurements of evolved hydrogen, biomass growth, and COD were performed. The concentration of carotenoides and bacteriochlorophyll was tested. In all tests the highest yield of hydrogen was obtained applying sodium glass photobioreactors illuminated with of 5 klx. Application of borosilicate glass or polycarbonate reactors reduced the amount of evolved hydrogen by 75 %. The best yield of hydrogen (2.1 dm 3 of H2 per dm 3 of medium) was obtained after illumination with 5 klx.

Microbial Biohydrogen Production by Rhodobacter sphaeroides O.U.001 in Photobioreactor

Pure strains of Purple Non-Sulfur (PNS) bacteria: Rhodobacter sphaeroides strain O.U 001 was studied for its efficiency to produce biohydrogen using the photofermentative pathways. An annular photobioreactor was being designed aiming the uniform light distribution. In prototype annular photobioreactor, at an initial pH of 6.8±0.2, temperature 32 ± 2 ◦C, inoculum volume 10% (v/v), inoculum age o...

Maximizing the solar to H2 energy conversion efficiency of outdoor photobioreactors using mixed cultures

A numerical study is presented aiming to maximize the solar to hydrogen energy conversion efficiency of a mixed culture containing microorganisms with different radiation characteristics. The green algae Chlamydomonas reinhardtii CC125 and the purple non-sulfur bacteria Rhodobacter sphearoides ATCC 49419 are chosen for illustration purposes. The previously measured radiation characteristics of ...

Improving Photobioreactor wall Using Optical Brightener: Investigating the Photostability of Coated Layer and Algal Growth

In this work, photostability, absorption and emission intensity of coated polycarbonate (PC) sheets with optical brightener (OB) as a wavelength converter material have been investigated. In addition, this coated sheet was used as a wall for microalgae culture flask, as a small scale photobioreactor, for studying the spectral conversion of UV-A radiation to blue light region and its effect ...

Materials, geometry, and net energy ratio of tubular photobioreactors for microalgal hydrogen production

We estimate the energy content, the operational energy inputs, and the net energy ratio (NER) of an industrial tubular photobioreactor used for the photosynthetic production of H2 by microalgae. The calculated H2 output of the photobioreactor is based on a range of algal photosynthetic H2 generation efficiencies, and on the application of standard theory for tubular solar collectors. Small diam...

Heterologous expression of glutamyl-tRNA reductase gene in Rhodobacter sphaeroides O.U.001 to enhance 5-aminolevulinic acid production

The pathways for synthesis of 5-aminolevulinic acid (5-ALA) use either succinyl-CoA and glycine (C-4 pathway), or glutamate (C-5 pathway). Although Rhodobacter sphaeroides synthesizes 5-ALA through the C-4 pathway, it also has the genes coding for the enzymes of the C-5 pathway, except for glutamyl-tRNA reductase. The glutamyl-tRNA reductase gene was cloned from Rhodospirillum rubrum and expres...
